IOA-244 + avelumab is an **experimental combination therapy** consisting of IOA-244, a first-in-class, highly selective, non–ATP-competitive phosphoinositide 3-kinase delta (PI3Kδ) inhibitor, and avelumab, an anti–PD-L1 monoclonal antibody[3][6][4]. IOA-244 is developed to modulate the tumor microenvironment and enhance antitumor immune responses by inhibiting regulatory T cell proliferation while favoring memory-like CD8+ T cell differentiation. Avelumab blocks the PD-L1/PD-1 interaction, thereby relieving immune inhibition and promoting antitumor immunity. The combination is under investigation in Phase 1b/2 trials for advanced/metastatic solid tumors, including melanoma and non-small cell lung cancer (NSCLC), aiming to increase the anti-tumor immune response.
