IOA-244 + pemetrexed + cisplatin + avelumab is an investigational multi-agent combination regimen used in clinical trials for the treatment of solid tumors. IOA-244 is a first-in-class, non–ATP-competitive, highly selective phosphoinositide 3-kinase delta (PI3Kδ) inhibitor with direct antitumor activity and immune-modulatory effects[1][5]. Pemetrexed is an antifolate small molecule chemotherapeutic agent, and cisplatin is a platinum-based DNA cross-linking chemotherapy; both are frequently co-administered to treat several solid tumors[2][4][6]. Avelumab is a human monoclonal antibody targeting programmed cell death protein 1 ligand 1 (PD-L1) and acts as an immune checkpoint inhibitor (anti-PD-L1 immunotherapy). This regimen leverages IOA-244’s Treg-inhibiting and anti-tumor properties to sensitize tumors to immunotherapy, the cytotoxic effects of pemetrexed and cisplatin, and the immune-stimulating effects of avelumab[1][5]. Clinical trials investigate this combination in malignant pleural mesothelioma, non-small cell lung cancer, and other solid and hematologic tumors[3].
