Iodine-125-labeled monoclonal antibody 425 is a radiolabeled therapeutic agent consisting of the murine monoclonal antibody MAb 425 conjugated with the radioactive isotope iodine-125. This drug specifically targets and binds to the epidermal growth factor receptor (EGFR) expressed on tumor cells. Upon binding, it is internalized by EGFR-expressing cells and delivers localized cytotoxic gamma radiation directly to malignant tissue. The primary clinical application has been as an adjuvant treatment for high-grade brain gliomas such as glioblastoma multiforme and astrocytoma with anaplastic foci. Clinical trials have demonstrated increased median survival in patients treated with this agent following surgery and external beam radiation therapy[1][2][5].
