Overview

Iodine 125-labeled monoclonal antibody A33 is a radiolabeled murine IgG2a monoclonal antibody that targets the A33 antigen, a cell-surface glycoprotein expressed in over 95% of human colon carcinomas and normal colon epithelium. The drug is designed for radioimmunotherapy and diagnostic imaging in colorectal cancer. After binding to the target antigen on tumor cells, the radiolabeled antibody is internalized into cytoplasmic vesicles within the cell. The radioactive iodine-125 delivers localized radiation to tumor cells, resulting in cytotoxicity while sparing most normal tissues due to selective targeting. Clinical studies have shown specific localization and prolonged retention of the isotope at tumor sites with minimal toxicity at tested doses[1][4][5][6].
