Overview

Iodine 131-labeled monoclonal antibody A33 is a radiopharmaceutical composed of the murine or humanized monoclonal antibody A33 conjugated to radioactive iodine-131. The mAb targets the A33 antigen, a cell surface glycoprotein homogeneously expressed in over 95% of human colon cancers and normal colon cells but not secreted or shed. Upon binding to the antigen on tumor cells, a fraction of membrane-bound mAb is internalized into endosomes. The drug delivers targeted beta radiation to tumor tissue via its radioactive payload (iodine-131), resulting in localized cytotoxicity and antitumor effects. It has been investigated primarily for advanced colorectal cancer patients who have failed conventional chemotherapy[1][2][3][6]. Preclinical and early clinical studies demonstrated specific tumor targeting and modest antitumor activity.
