Iodine-131 tenatumomab is a radiolabeled monoclonal antibody drug composed of the anti-tumor monoclonal antibody tenatumomab conjugated to the radioisotope iodine-131. This agent is developed for targeted radioimmunotherapy, aiming to deliver cytotoxic ionizing radiation directly to tumor cells expressing the target antigen recognized by tenatumomab. The conjugation of iodine-131 enables the antibody to deliver beta emissions that cause DNA damage and cell death in targeted cancer cells, while the monoclonal antibody component confers antigen specificity, leading to more selective tumor cell cytotoxicity and minimizing effects on non-target cells. Tenatumomab itself targets the A1 antigen expressed in certain tumors. The radiolabeled antibody is generally administered by intravenous infusion and is intended for the treatment of solid tumors or hematologic malignancies that express the relevant antigen.
