Iodine i 131 CC49-DELTACH2 monoclonal antibody is a humanized monoclonal antibody engineered to target the tumor-associated glycoprotein 72 (TAG-72) antigen, which is overexpressed in many gastrointestinal and other adenocarcinomas. The DELETED CH2 (delta CH2) modification improves pharmacokinetics and tumor penetration by enhancing blood clearance. Labeled with the radioisotope iodine-131, this agent selectively binds to TAG-72-positive cancer cells and delivers cytotoxic radiation directly to tumors, limiting damage to normal tissues. The mechanism leverages both the antibody-antigen specificity and targeted radiotherapy (radioimmunotherapy). It is primarily under evaluation for gastrointestinal cancers and other TAG-72–expressing tumors, with phase I clinical studies indicating tumor localization and grade IV myelosuppression as the main toxicity[1][2][4][7][8].
