Overview

Iodine I-131 labetuzumab is a radioimmunotherapy (RAIT) agent comprising the humanized monoclonal antibody labetuzumab (hMN-14) labeled with the radioactive isotope iodine-131. Labetuzumab specifically targets the carcinoembryonic antigen-related cell adhesion molecule 5 (CEACAM5, or CEA), which is highly expressed in various epithelial tumors, including colorectal, gastrointestinal, and medullary thyroid cancers. Upon intravenous administration, the antibody localizes to tumor sites, where the iodine-131 component emits beta particles that induce lethal DNA damage in cancer cells. Additionally, the gamma emissions from iodine-131 allow for scintigraphic imaging to monitor drug biodistribution. Developed by Immunomedics (now part of Gilead Sciences), the agent was primarily investigated as an adjuvant treatment following the resection of colorectal liver metastases and for managing metastatic medullary thyroid carcinoma.
