Iodine I 131 monoclonal antibody 81C6 is a radioimmunoconjugate composed of the murine IgG2 anti-tenascin monoclonal antibody (clone: 81C6) covalently labeled with the radioactive isotope iodine‑131. The drug specifically targets tenascin, an extracellular matrix protein that is highly overexpressed in gliomas and some other cancers. Upon binding to tenascin on tumor cells, the conjugated radioactive isotope delivers localized beta radiation directly to malignant tissue while sparing surrounding healthy brain tissue. This targeted approach aims to improve outcomes for patients with high-grade brain tumors such as glioblastoma multiforme (GBM). The agent has been investigated under the brand name Neuradiab in clinical trials for recurrent or newly diagnosed GBM and other brain neoplasms. Although early-phase studies showed promising results regarding progression-free survival and safety profile, a pivotal phase III trial was terminated due to logistical issues before efficacy could be definitively established[1][2][3][4][5].
