iODN2088 is a synthetic inhibitory oligodeoxynucleotide (iODN) that functions as a potent and specific antagonist of Toll-like receptor 9 (TLR9). It consists of a phosphorothioate-modified sequence that includes a G-rich inhibitory motif, which allows it to competitively bind to TLR9 and prevent the recognition of stimulatory CpG-containing DNA. By blocking TLR9 activation, iODN2088 suppresses the recruitment of MyD88 and the subsequent activation of the NF-κB and MAPK signaling pathways, thereby reducing the production of pro-inflammatory cytokines. While primarily utilized as a research tool to study the role of TLR9 in innate immunity and inflammatory pathologies such as atherosclerosis and autoimmune diseases, it represents a class of molecules with potential therapeutic applications in modulating aberrant immune responses.
