Overview

**Iododiflunisal** is a small-molecule derivative of diflunisal that acts as a potent stabilizer of the transthyretin (TTR) tetramer and functions as a molecular chaperone of the TTR-amyloid beta (Aβ) interaction[3][11][12][14]. It is being investigated for its potential to prevent or delay amyloid fibril formation, particularly in the context of transthyretin amyloidosis and Alzheimer's disease. Iododiflunisal binds deeply within the transthyretin hormone-binding channel, where its iodine and fluorine substituents provide highly specific and strong interactions, resulting in significant stabilization of the TTR tetramer and inhibiting amyloidogenesis[12]. Preclinical studies indicate oral administration of iododiflunisal improves Alzheimer's disease-like pathology in animal models[3][11][14]. Its mechanism is primarily based on stabilization of TTR, blocking its dissociation and subsequent aggregation into amyloid fibrils, which underlies certain forms of amyloidosis[11][12][14]. Iododiflunisal is also radiolabeled for PET imaging applications in experimental settings[3].
