Ioflubenzamide I-131 is an iodine 131-radiolabeled small-molecule benzamide compound developed for the targeted treatment of metastatic melanoma. The drug consists of a benzamide moiety that binds specifically to melanin, a pigment overexpressed in many melanomas. By binding to melanin, ioflubenzamide I-131 selectively delivers cytotoxic doses of gamma and beta radiation directly to melanin-expressing tumor cells, sparing non-melanin tissues. Preclinical studies demonstrated significant antitumor activity in human melanoma xenograft models, with multiple doses resulting in durable tumor regression. Clinical development reached Phase 1 but was terminated for unknown reasons[2][3][4].
