ION537 is a generation 2.5 antisense oligonucleotide designed to inhibit the expression of Yes-associated protein 1 (YAP1), a transcription factor implicated in tumor cell survival and resistance mechanisms in various cancers. Developed by Ionis Pharmaceuticals in collaboration with the University of Texas MD Anderson Cancer Center, it employs RNA interference to reduce YAP1 mRNA levels, thereby hindering YAP-driven oncogenic processes. Preclinical studies have shown that targeting YAP1 can be particularly effective in certain hepatocellular and head and neck cancers, as well as other solid tumors where YAP1 plays a critical role. The drug has completed Phase 1 clinical trials for advanced solid tumors but has not progressed beyond this stage as of early 2024[1][2][3][4][5][6].
