IONC101 is a systemically delivered, mRNA-based immunotherapy developed by iOncologi for the treatment of various solid tumors, including adult and pediatric glioblastoma, pediatric osteosarcoma, and head and neck squamous cell carcinoma (HNSCC). The drug utilizes proprietary Multilamellar Lipid Particle Aggregate (MLPA) technology to deliver mRNA encoding a rational combination of immune-modulating proteins. These proteins are designed to reprogram both innate and adaptive immune components by driving effector T cell recruitment, reversing myeloid-derived suppressor cell (MDSC) activity, and inducing local inflammation within the tumor microenvironment. By targeting both tumor-derived and off-the-shelf antigens, IONC101 aims to elicit a rapid, broad, and long-lasting anti-tumor immune response while overcoming tumor-induced immunosuppression and tolerance.
