Overview

IONIS-FGFR4Rx is an antisense oligonucleotide drug developed to target and inhibit fibroblast growth factor receptor 4 (FGFR4). Its mechanism of action involves antagonizing FGFR4, a receptor implicated in metabolic regulation, particularly in liver and adipose tissues. The drug was primarily investigated for the treatment of obesity, aiming to increase metabolism and fat burning while improving insulin sensitivity. Developed by Ionis Pharmaceuticals (formerly Isis Pharmaceuticals), with origins at the University of Queensland, it reached Phase 2 clinical trials for obesity but development was discontinued after this stage[3][5][6]. Preclinical studies indicated reductions in body fat and reversal of existing obesity without central nervous system side effects[5].
