Overview

IOWH-032 is a synthetic small molecule that acts as a potent inhibitor of the cystic fibrosis transmembrane conductance regulator (CFTR) chloride channel. It was developed as an anti-secretory and anti-diarrheal agent for the treatment of cholera and other forms of secretory diarrhea. The compound inhibits CFTR-mediated chloride secretion in intestinal epithelial cells, thereby reducing water loss associated with diarrheal diseases. In preclinical models and clinical trials up to Phase 2a, IOWH-032 demonstrated safety but did not show statistically significant efficacy in reducing stool output compared to placebo in cholera patients. The molecule features an oxadiazole-carboxamide core with a dibromo-hydroxyphenyl pharmacophore[1][2][3][4][5][6][8].
