Overview

Ipalolitovorelimab (KN046) is a first-in-class bispecific monoclonal antibody designed to simultaneously target two key immune checkpoints: Programmed Death-Ligand 1 (PD-L1) and Cytotoxic T-Lymphocyte-Associated Antigen 4 (CTLA-4). Developed by Alphamab Oncology using a proprietary heterodimerization platform, the drug aims to enhance T-cell activation and overcome tumor-induced immunosuppression more effectively than single-agent checkpoint inhibitors. By blocking both the PD-L1/PD-1 and CTLA-4/B7 pathways, ipalolitovorelimab restores anti-tumor immune responses across a variety of solid tumors. It is currently undergoing evaluation in late-stage clinical trials for indications including non-small cell lung cancer (NSCLC), pancreatic cancer, and cervical cancer. In locally advanced cervical cancer, it is being investigated in combination with neoadjuvant chemotherapy to improve pathological complete response rates and long-term survival outcomes.
