Ipamorelin is a synthetic pentapeptide and selective agonist of the ghrelin/growth hormone secretagogue receptor (GHSR, also known as GHSR-1a). It mimics the action of ghrelin by binding to GHSR in the brain and other tissues, thereby selectively stimulating the release of growth hormone (GH) from the pituitary gland[1][3][4][8]. Unlike some other GH secretagogues, ipamorelin does not significantly affect levels of prolactin, follicle-stimulating hormone (FSH), luteinizing hormone (LH), thyroid-stimulating hormone (TSH), adrenocorticotropic hormone (ACTH), or cortisol[4]. Its selectivity for GH release makes it distinct among similar peptides. Ipamorelin was originally developed by Novo Nordisk and later investigated by Helsinn Therapeutics for postoperative ileus but development was discontinued due to lack of efficacy[4]. It has also been used off-label in performance enhancement settings.
