Overview

Iparomlimab + tuvonralimab + lenvatinib is a combination therapy being investigated for the neoadjuvant treatment of hepatocellular carcinoma (HCC). This regimen combines two bispecific antibodies developed by Akeso Biopharma with a multi-kinase inhibitor. **Iparomlimab** (cadonilimab) is a first-in-class bispecific antibody targeting programmed cell death protein 1 (PD-1) and cytotoxic T-lymphocyte-associated protein 4 (CTLA-4), designed to enhance anti-tumor immune responses while potentially reducing the toxicity associated with combining separate PD-1 and CTLA-4 inhibitors. **Tuvonralimab** (ivonescimab) is a bispecific antibody targeting PD-1 and vascular endothelial growth factor (VEGF), intended to simultaneously block immune escape and inhibit tumor angiogenesis. **Lenvatinib** is an oral receptor tyrosine kinase inhibitor that targets VEGF receptors (VEGFR1-3), fibroblast growth factor receptors (FGFR1-4), PDGFRα, KIT, and RET. The combination aims to leverage synergistic effects by targeting multiple pathways involved in tumor growth, immune evasion, and vascularization.
