iparomlimab + tuvonralimab + TFOX

This is a combination therapy consisting of QL1706 (a dual immune checkpoint inhibitor containing iparomlimab, an anti-PD-1 antibody, and tuvonralimab, an anti-CTLA-4 antibody) and the TFOX chemotherapy regimen (docetaxel, oxaliplatin, leucovorin, and 5-fluorouracil). Developed by Qilu Pharmaceutical (QL1706 component) and being studied by The First Affiliated Hospital with Nanjing Medical University, this regimen is intended as a first-line treatment for patients with HER2-negative advanced gastric or gastroesophageal junction (GEJ) adenocarcinoma. QL1706 utilizes a unique Mab-Pair technology to produce two distinct antibodies from a single cell line, targeting both PD-1 and CTLA-4 pathways to enhance anti-tumor immune responses. The TFOX regimen, a modified version of the FLOT chemotherapy protocol, provides direct cytotoxic activity and potentially increases tumor immunogenicity to synergize with the dual checkpoint blockade.
