Overview

This four-drug combination is being studied as a neoadjuvant therapy for patients with locally advanced triple-negative breast cancer (TNBC). The therapy seeks to exploit the synergistic antitumor activity of targeted AKT inhibition, immune checkpoint blockade, and cytotoxic chemotherapy, potentially offering enhanced efficacy for high-risk breast cancer. Ipatasertib inhibits the PI3K/AKT pathway, atezolizumab blocks the PD-L1/PD-1 axis to release immune suppression, and both doxorubicin and cyclophosphamide provide direct cytotoxic activity against tumor cells[5][6].
