Overview

IPG0521 is a monoclonal antibody antagonist targeting C-C motif chemokine receptor 8 (CCR8), developed by Nanjing Immunophage Biotech for the treatment of solid tumors and IgG4-related diseases. Unlike many competing CCR8 antibodies that rely on potent antibody-dependent cellular cytotoxicity (ADCC) to deplete regulatory T cells (Tregs), IPG0521 primarily functions by blocking the chemotaxis and immunosuppressive activity of CCR8+ Tregs within the tumor microenvironment. It possesses only mild ADCC activity, which may offer a safety advantage by avoiding systemic Treg depletion. Preclinical data indicates high-affinity cross-reactivity across five species (human, monkey, dog, rat, and mouse) and synergistic effects when combined with anti-PD-1 therapies in models of lung, liver, and breast cancers.
