IPG11406 is a highly potent small molecule antagonist of G protein-coupled receptor 183 (GPR183), also known as Epstein-Barr virus-induced gene 2 (EBI2). Developed by Nanjing Immunophage Biotech, it is designed to suppress EBI2-mediated activation and chemotaxis of inflammatory cells such as dendritic cells, macrophages, T cells, and B cells. By blocking this pathway, IPG11406 aims to resolve inflammation and reverse autoimmunity in several autoimmune disease models. Preclinical studies have shown efficacy in models for rheumatoid arthritis, systemic lupus erythematosus (SLE), multiple sclerosis (MS), and inflammatory bowel disease (IBD). The drug has demonstrated good aqueous solubility, selectivity for its target, favorable pharmacokinetics, and an excellent safety profile in early clinical trials. It is currently being evaluated in phase IIa clinical trials for lupus nephritis and has completed phase I studies for SLE/LN as well as other autoimmune diseases[1][2][3][4][5].
