Overview

IPG1576 is a small molecule inhibitor of Macrophage Migration Inhibitory Factor (MIF), a pleiotropic pro-inflammatory cytokine that plays a significant role in promoting an immunosuppressive tumor microenvironment. MIF is often overexpressed in various malignancies, where it contributes to the recruitment and polarization of M2-like macrophages and the induction of T-cell exhaustion. Preclinical research presented at the 2025 World Conference on Lung Cancer (WCLC) indicates that IPG1576 can reshape the immune landscape in models of pulmonary sarcomatoid carcinoma (PSC), a rare and aggressive subset of non-small cell lung cancer. By inhibiting MIF, IPG1576 aims to reverse resistance to PD-1/PD-L1 checkpoint inhibitors, particularly when used in combination with other microenvironment-modulating agents such as CCR1 or CCR5 antagonists.
