IPH43 is a preclinical **antibody-drug conjugate** being developed by **Innate Pharma** in collaboration with **AstraZeneca** for oncology. It consists of a glycosylated anti-MICA monoclonal antibody linked site-specifically to a **pyrrolobenzodiazepine dimer** cytotoxic payload, producing a homogeneous ADC with a drug-to-antibody ratio close to 2. IPH43 is designed to bind **MHC class I polypeptide-related sequence A and B** on tumor cells, deliver a DNA-crosslinking payload to kill those cells, and also disrupt MICA/B interaction with **NKG2D** to help restore NK cell-mediated immune surveillance. The program has been described as being in **IND-enabling preclinical development** and is relevant across multiple MICA/B-expressing solid tumors and hematologic malignancies.
