IPH4502 is a novel and differentiated topoisomerase I inhibitor Antibody Drug Conjugate (ADC) conjugated to exatecan, targeting Nectin-4, a cell adhesion molecule overexpressed in several types of solid tumors including urothelial carcinoma, breast cancer, non-small cell lung cancer, and gastrointestinal cancers. The drug is designed to deliver the cytotoxic payload exatecan specifically to tumor cells expressing Nectin-4. Preclinical data have shown superior anti-tumor activity compared to enfortumab vedotin (EV), especially in models with low or heterogeneous Nectin-4 expression and in EV-resistant tumors. IPH4502 also demonstrates strong bystander killing effects and efficient internalization, supporting its potential for broad clinical application across multiple tumor types such as triple-negative breast cancer, head and neck squamous cell carcinoma, and esophageal cancer[1][6][8].
