IPH5401 + durvalumab is an investigational combination therapy for cancer immunotherapy. IPH5401 is a first-in-class monoclonal antibody targeting the C5a receptor (C5aR) expressed on subsets of myeloid-derived suppressor cells (MDSCs) and neutrophils in the tumor microenvironment. By blocking C5aR, IPH5401 aims to reduce immunosuppression within tumors. Durvalumab is a monoclonal antibody that blocks programmed death-ligand 1 (PD-L1), thereby enhancing immune system recognition and destruction of cancer cells. The combination seeks to synergistically enhance anti-tumor immunity by both reducing immune suppression (via C5aR blockade) and promoting T-cell activation (via PD-L1 inhibition). This approach is being evaluated primarily in solid tumors, including non-small cell lung cancer with secondary resistance to prior immuno-oncology treatment and IO-naïve hepatocellular carcinoma[5].
