IPI-459 is a small molecule inhibitor of the gamma isoform of phosphoinositide 3-kinase (PI3Kγ). Developed by Infinity Pharmaceuticals, it is primarily utilized in preclinical research to investigate the role of PI3K signaling in immune cell modulation and metabolic fitness. In the context of immuno-oncology, IPI-459 has been studied for its ability to enhance adoptive T cell therapy (ACT). Research indicates that inhibiting PI3Kγ during the priming or expansion of tumor-specific CD8+ T cells can prevent premature differentiation and improve mitochondrial function, thereby promoting more durable anti-tumor immunity and survival in models of solid tumors such as melanoma. While it shares a target with the clinical-stage candidate eganelisib (IPI-549), IPI-459 serves as a specific tool for exploring the therapeutic potential of selective PI3Kγ blockade.
