Overview

IPI-609 is a small molecule inhibitor of the Hedgehog (Hh) signaling pathway, specifically targeting the transmembrane protein Smoothened (SMO). Developed by Infinity Pharmaceuticals, IPI-609 is a semi-synthetic analog of the plant-derived alkaloid cyclopamine, engineered for improved potency and pharmaceutical properties. By binding to SMO, IPI-609 prevents the activation of downstream Gli transcription factors, thereby inhibiting the growth of tumors that are dependent on aberrant Hedgehog signaling. It has been studied preclinically in various malignancies, including basal cell carcinoma, medulloblastoma, and pancreatic cancer, where the Hh pathway plays a critical role in tumor initiation, progression, and the maintenance of the tumor microenvironment.
