Ipilimumab + degarelix is a combination therapy being investigated by Memorial Sloan Kettering Cancer Center for the treatment of early-stage prostate cancer. Ipilimumab is a fully human monoclonal antibody that inhibits Cytotoxic T-Lymphocyte Antigen 4 (CTLA-4), a checkpoint molecule that normally suppresses T-cell activation; by blocking CTLA-4, ipilimumab enhances the immune system's ability to recognize and destroy cancer cells. Degarelix is a synthetic peptide and gonadotropin-releasing hormone (GnRH) receptor antagonist that rapidly suppresses testosterone production, a key driver of prostate cancer growth. The combination is specifically being evaluated in a Phase 2 clinical trial (NCT02020070) for men with newly diagnosed metastatic castration-sensitive prostate cancer (mCSPC) or biochemically recurrent prostate cancer, often in conjunction with radical prostatectomy, to determine if early intervention with dual hormonal and immune therapy can improve clinical outcomes before the disease progresses to a castration-resistant state.
