ipilimumab + LTX-315 is a combination therapy consisting of the immune checkpoint inhibitor ipilimumab and the oncolytic peptide LTX-315. Ipilimumab is a monoclonal antibody that targets cytotoxic T-lymphocyte-associated protein 4 (CTLA4), blocking its inhibitory effect on T cells and thereby enhancing antitumor immune responses. LTX-315 is a first-in-class, 9-mer cationic oncolytic peptide derived from bovine lactoferrin, designed for intratumoral injection. It induces immunogenic cell death by disrupting tumor cell membranes and mitochondria, leading to the release of damage-associated molecular patterns that promote local and systemic antitumor immunity. The combination aims to synergistically enhance T-cell infiltration into tumors and improve clinical outcomes in patients with advanced solid tumors such as malignant melanoma. Clinical studies have shown this regimen to be generally safe with manageable toxicity profiles, increased CD8+ T-cell infiltration in treated lesions, clonal expansion of tumor-reactive T cells in blood and tumor tissue, and evidence of disease stabilization in some patients[3][5][6].
