Overview

This combination therapy protocol, evaluated in a Phase II clinical trial (NCT03472586) at the Sidney Kimmel Cancer Center at Thomas Jefferson University, is designed for patients with metastatic uveal melanoma, particularly those with liver metastases. The regimen combines two systemic immune checkpoint inhibitors, ipilimumab (a CTLA-4 inhibitor) and nivolumab (a PD-1 inhibitor), with a localized liver-directed procedure called immunoembolization. During immunoembolization, granulocyte-macrophage colony-stimulating factor (GM-CSF) is infused into the hepatic artery, followed by the administration of an embolic agent to obstruct blood flow to the tumor. This dual approach aims to induce tumor cell death through ischemia while simultaneously stimulating a robust local and systemic immune response against the cancer cells.
