Overview

Ipilimumab + nivolumab + pembrolizumab is a combination of three immune checkpoint inhibitor monoclonal antibodies used in cancer immunotherapy. Ipilimumab targets cytotoxic T-lymphocyte-associated protein 4 (CTLA-4), while both nivolumab and pembrolizumab target programmed cell death protein 1 (PD-1). By blocking these inhibitory pathways, the combination enhances T-cell mediated immune responses against tumor cells. This regimen is considered for patients with advanced cancers, particularly after progression on prior immunotherapies, to potentially overcome resistance and improve antitumor activity[2][4]. Each component has been individually approved for multiple cancer types; however, the triple combination is primarily investigated in clinical trials or off-label settings.
