Overview

A triple combination immunotherapy regimen consisting of ipilimumab (CTLA-4 inhibitor), nivolumab (PD-1 inhibitor), and talimogene laherparepvec (T-VEC, an oncolytic virus). This combination leverages three different immunotherapeutic mechanisms to potentially enhance antitumor activity in advanced melanoma. T-VEC selectively replicates in and lyses tumor cells, releasing tumor-derived antigens and producing GM-CSF locally. Ipilimumab enhances T-cell recruitment and prevents exhaustion of activated T cells. Nivolumab further enhances antitumor immunity by blocking the PD-1/PD-L1 pathway.
