All are monoclonal antibodies used in cancer immunotherapy. - **Ipilimumab** targets cytotoxic T lymphocyte antigen 4 (CTLA-4), blocking an inhibitory signal to T cells and enhancing immune response against tumor cells[9][1]. - **Pembrolizumab** is a humanized IgG4 monoclonal antibody that binds to programmed cell death protein 1 (PD-1) on T cells, preventing its interaction with PD-L1/PD-L2 and thereby promoting anti-tumor immunity[1][2]. - **Durvalumab** is a human IgG1 kappa monoclonal antibody that binds to programmed death-ligand 1 (PD-L1), inhibiting its interaction with PD-1 and CD80, thus restoring immune responses against cancer cells[4][6]. This triple combination has not been approved as a single regimen but represents an investigational approach leveraging multiple checkpoint blockades for potentially synergistic effects in advanced cancers.
