Ipiwutide (also known as Epivotide, ipiluvitide, or TAT-N24) is a synthetic peptide inhibitor targeting the p55 regulatory subunit of phosphatidylinositol 3-kinase (p55PIK, encoded by the *PIK3R3* gene). Developed by Wuhan Yicheng Biotechnology, ipiwutide acts as a broad-spectrum anti-inflammatory agent by specifically blocking the interaction of p55PIK with its binding partners, such as the retinoblastoma protein (Rb), thereby inhibiting cell cycle progression and downstream NF-κB activation. This mechanism suppresses the production and secretion of key inflammatory cytokines (including TNF-α, IL-1, IL-2, and IL-6) and reduces inflammatory cell migration and infiltration. Ipiwutide is being developed in various formulations, including ophthalmic solutions, nasal drops, and nebulizer solutions, for the treatment of inflammatory and ocular conditions such as allergic conjunctivitis, dry eye syndrome, neurotrophic keratitis, allergic rhinitis, and post-cataract surgery inflammation.
