**IPN60090 + pembrolizumab** is a combination therapy of a novel, selective, oral glutaminase-1 (GLS1) inhibitor (IPN60090, also known as IACS-6274) with the anti-PD-1 immune checkpoint inhibitor pembrolizumab (marketed as Keytruda). IPN60090 (IACS-6274) targets cancer cell metabolism by inhibiting glutaminase-1, thereby impairing glutamine-dependent tumor proliferation. Pembrolizumab is a monoclonal antibody that targets the programmed cell death protein 1 (PD-1) receptor, blocking its interaction with PD-L1 and PD-L2, which enhances T cell–mediated antitumor immune responses. This combination aims to exploit metabolic vulnerabilities in tumors while simultaneously activating antitumor immunity. Early-phase trials focus on advanced solid tumors, particularly those with metabolic or immune biomarker selection.[1][2][4]
