IPN60340 + azacitidine + venetoclax is an investigational triplet combination therapy being developed by Ipsen for the treatment of newly diagnosed acute myeloid leukemia (AML) in patients who are ineligible for intensive chemotherapy. The regimen consists of **IPN60340**, a first-in-class molecular glue degrader of **GSPT1** (G1 to S phase transition 1), combined with the standard-of-care agents **azacitidine** (a hypomethylating agent) and **venetoclax** (a BCL-2 inhibitor). IPN60340, which originated from a collaboration between Ipsen and Plexium, induces the proteasomal degradation of GSPT1, a translation termination factor essential for the survival of AML cells. By combining this novel mechanism with DNA methyltransferase inhibition and BCL-2-mediated apoptosis induction, the triplet aims to overcome resistance and improve remission rates compared to the doublet of azacitidine and venetoclax. The combination is currently being evaluated in a global Phase 2b/3 randomized, double-blind study (NCT06340100).
