Overview

iPSC-Derived CD8 αβ T Cells are an adoptive cell therapy derived from inducible pluripotent stem cells (iPSCs). These engineered CD8αβ T cells express a T-cell receptor (TCR) specifically targeting HLA-A24-restricted GPC3 peptides, enabling TCR-dependent killing of tumor cells. Additionally, they are designed for innate, TCR-independent killing through markers like NKG2D and DNAM1. The cells are armored with constitutive IL-15/IL-21 secretion to enhance acute and serial killing activity and include immune evasion edits to prevent cellular and humoral rejection, aiming for enhanced persistence in the tumor microenvironment. This therapy is being developed to address challenges in solid tumors, such as target antigen downregulation and cell trafficking.
