Drug intelligence / Profile preview
IQ-1 is a cell-permeable small-molecule modulator of the Wnt/β-catenin signaling pathway. It acts by binding to the PR72/130 (PPP2R3A) regulatory subunit of protein phosphatase 2A (PP2A), which blocks the interaction between PP2A and Naked cuticle (Nkd). This blockade prevents the PP2A-mediated dephosphorylation of the coactivator p300 (EP300) at Ser89, thereby disrupting the β-catenin/p300 interaction and preventing the switch of β-catenin coactivator usage from CBP (CREBBP) to p300. Consequently, IQ-1 selectively enhances β-catenin/CBP-mediated transcription at the expense of β-catenin/p300-mediated transcription. This mechanism is widely used in stem cell research to maintain the pluripotency and self-renewal of embryonic stem cells (ESCs) in vitro, and has been investigated for its ability to rescue hematopoietic stem cell (HSC) repopulating activity.
