Drug intelligence / Profile preview
Ir-MT1 is a mitochondria-targeted iridium(III)-based photosensitizer developed for the photoimmunotherapy of renal cell carcinoma (RCC). Upon photoactivation with light, Ir-MT1 generates high levels of reactive oxygen species (ROS) within the mitochondria, leading to mitochondrial DNA leakage and calcium homeostasis disruption. This triggers a cascade of events including Caspase-1 cleavage and activation of the cGAS-STING and RIPK1-MLKL pathways, driving synergistic pyroptosis and necroptosis. This multi-pathway cell death promotes immunogenic cell death (ICD), enhancing dendritic cell activation, T-cell infiltration, and long-lasting antitumor immune memory to prevent tumor recurrence and metastasis.
