Overview

IRAK1 shRNA is a gene-silencing therapeutic candidate designed to knock down the expression of interleukin-1 receptor-associated kinase 1 (IRAK1). IRAK1 is a critical mediator in the Toll-like receptor (TLR) and Interleukin-1 receptor (IL-1R) signaling pathways, which are highly activated in cisplatin-resistant ovarian cancer and cancer stem cell (CSC) populations. By utilizing short hairpin RNA (shRNA) to reduce IRAK1 levels, this approach aims to impair CSC-enriched spheroid growth and orthotopic tumor progression. Research conducted at the University of Kansas Medical Center indicates that IRAK1 knockdown leads to the downregulation of stemness-related genes, including STAT3, MYC, and NOTCH, as well as a reduction in the cisplatin efflux transporter ABCC1. This modality is being explored as a potential strategy to overcome chemoresistance and recurrence in advanced ovarian cancer.
