iratumumab

Iratumumab is a fully human IgG1-kappa monoclonal antibody that targets CD30, a member of the tumor necrosis factor receptor superfamily expressed on activated lymphocytes and certain lymphoma cells. Developed originally by Medarex (later acquired by Bristol Myers Squibb), it was investigated for the treatment of CD30-positive lymphomas, including Hodgkin's disease and anaplastic large cell lymphoma. The mechanism of action involves binding to CD30 with high affinity, neutralizing its bioactivity by preventing interaction with its ligand (CD30L), thereby inhibiting downstream signaling pathways such as NF-kB and MAPK. This results in reduced proliferation of malignant cells and induction of antibody-dependent cytotoxicity. Iratumumab received orphan drug designation for Hodgkin's lymphoma but development was discontinued after phase II trials[1][3][6][7].
