IRBM-Z-2 is a small molecule antiviral compound developed by IRBM in collaboration with the CNCCS consortium. It acts as a potent allosteric inhibitor of the Zika virus (ZIKV) NS2B-NS3 protease, a critical enzyme for viral replication. The compound features a unique N-acylsydnone imine moiety that binds to a previously unidentified allosteric pocket, stabilizing the protease in a catalytically inactive state. Beyond ZIKV, IRBM-Z-2 exhibits inhibitory activity against other orthoflaviviruses, including dengue virus serotype 2 (DENV2) and West Nile virus (WNV). Preclinical evaluations demonstrated high oral bioavailability and significant therapeutic efficacy in mouse models of ZIKV infection, where it provided complete survival.
