Overview

IRDye800CW-anti-HER2 nanobody is an optical imaging agent composed of an anti-HER2 nanobody (single-domain antibody, also called VHH) site-specifically conjugated to the near-infrared fluorescent dye IRDye800CW. The agent is designed for rapid, high-contrast molecular imaging or fluorescence-guided surgery (FGS) of HER2-positive tumors, particularly breast cancer. Nanobodies, compared to monoclonal antibodies, are significantly smaller, have faster tumor penetration, and result in superior tumor-to-background imaging ratios shortly after administration. Mechanistically, the anti-HER2 nanobody binds with high affinity and specificity to the extracellular domain of the HER2 receptor (ErbB2), commonly overexpressed in certain cancers. The fluorescent label enables intraoperative visualization of tumor margins in real time. Preclinical studies demonstrate 10–20 fold faster tumor accumulation and superior contrast compared to trastuzumab-based probes, with binding preserved after conjugation, and rapid clearance from non-tumor tissues. The agent is designed for intravenous injection and enables real-time image-guided tumor resection[3][4].
