Irinosome High C is an optimized liposomal formulation of irinotecan, a semi-synthetic analogue of the natural alkaloid camptothecin. It is designed to improve the therapeutic index of irinotecan by enhancing its delivery to tumor sites and modulating the tumor immune microenvironment (TIME). Specifically, Irinosome High C is engineered to shift the TIME from an immunosuppressive state to an immunosupportive one, making it a potent candidate for combination therapies with immunomodulators like interleukin-12 (IL-12). In preclinical models of metastatic melanoma, it has demonstrated the ability to work synergistically with self-amplifying RNA (saRNA) lipid nanoparticles to reduce tumor burden and improve survival by stimulating a robust antitumor immune response involving cytokines such as IL-2 and IFN-γ.
