This is a combination of two small molecule drugs used in oncology research. Irinotecan is a topoisomerase I inhibitor that interferes with DNA replication and transcription, leading to cell death. Linsitinib (also known as OSI-906 or ASP7487) is an oral small molecule inhibitor targeting the insulin-like growth factor 1 receptor (IGF-1R) and the insulin receptor (IR), blocking signaling pathways involved in tumor growth and survival. The combination has been studied for safety and tolerability in patients with advanced cancers refractory to standard therapy. The rationale for combining these agents is to enhance antitumor efficacy by simultaneously inhibiting DNA repair mechanisms and growth factor signaling pathways[1][2][3][4].
