This combination refers to a sequential chemotherapy regimen consisting of FOLFOX (oxaliplatin, leucovorin, and 5-fluorouracil) and FOLFIRI (irinotecan, leucovorin, and 5-fluorouracil) evaluated by Dong-A University Hospital for the treatment of advanced or relapsed gastric cancer. Oxaliplatin is a third-generation platinum analogue that forms DNA adducts, inhibiting DNA synthesis. Irinotecan is a topoisomerase I inhibitor that prevents DNA religation, causing lethal double-strand breaks. 5-fluorouracil is an antimetabolite that inhibits thymidylate synthase, while leucovorin is used to enhance the cytotoxic effect of 5-fluorouracil. The regimen was studied in a Phase II trial (NCT01138904) to determine the optimal sequence of these therapies as first-line and second-line treatments in patients with metastatic or relapsed gastric cancer, adapting a strategy previously established in colorectal cancer.
