Overview

Irinotecan is a small molecule topoisomerase I inhibitor that prevents DNA religation, leading to double-strand breaks and cell death. Oxaliplatin is a platinum-based antineoplastic agent that forms DNA cross-links, inhibiting DNA replication and transcription. Cetuximab is a recombinant human/mouse chimeric monoclonal antibody that binds specifically to the extracellular domain of the epidermal growth factor receptor (EGFR), inhibiting downstream signaling pathways involved in cell proliferation and survival. This specific triple combination was evaluated in a Phase II clinical trial (NCT00871169) sponsored by the New Mexico Cancer Research Alliance for the treatment of locally advanced or metastatic pancreatic cancer.
