Overview

HAI chronomodulated chemotherapy is a regional combination chemotherapy regimen delivered via hepatic artery infusion (HAI) according to circadian rhythms (chronomodulation). The regimen typically consists of a triplet of small molecule agents: irinotecan, oxaliplatin, and 5-fluorouracil (5-FU). It is primarily developed by the Association pour la Recherche sur le Temps Biologique et la Chronothérapie (ARTBC) for the treatment of colorectal cancer with unresectable liver metastases. The chronomodulated delivery aims to synchronize drug administration with the body's biological clock to maximize efficacy and minimize toxicity. In clinical studies like the OPTILIV trial, this HAI triplet has been evaluated in combination with systemic cetuximab to improve surgical resection rates.
